Application of polymerase chain reaction-restriction fragment length polymorphism (RFLP-PCR) in the analysis of single nucleotide polymorphisms (SNPs)

Polymerase chain reaction-restriction fragment length polymorphism (RFLP-PCR) is a technique used to identify single nucleotide polymorphisms (SNPs) based on the recognition of restriction sites by enzymes. RFLP-PCR an easy-to-perform and inexpensive tool for initial analysis SNPs potentially associated with some monogenic diseases, as well in genotyping, genetic mapping, lineage screening, forensics ancient DNA analysis. The method employs four steps: (1) isolation material PCR; (2) digestion amplicons; (3) electrophoresis digested fragments; (4) visualisation. Despite its obsolescence presence high-throughput techniques, it still applied disease entities variation severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). low-cost low-throughput research allowing absence specialised equipment, useful when there limited budget.
